The Fairfax Lab is the back cover story of the March 2019 issue of the European Journal of Immunology. The back cover features a confocal tile scan of popliteal lymph nodes from 4getIL‐4R−/− mice at steady‐state stained for B220 (Red), CD4 (Blue), and FDC‐M2 (Grey). B‐cell follicles are disorganized, with B‐cells clustering around aberrantly positioned follicular dendritic cells. The image is taken from Cortes‐Selva et al. (pp. 428–442), where the authors show that the absence of IL‐4 signaling leads to mal‐positioning of both hematopoietic and stromal cells. Immunization of these mice with Type‐2 antigens fails to induce lymph node reorganization or germinal center formation, while Type‐1 antigen induced reorganization in the absence of IL‐4 is IFNγ dependent. These results demonstrate a novel role of IL‐4 in peripheral lymph node architecture during both homeostasis and antigenic challenge.
